ETR028 is a novel hydrocodone prodrug developed by Elysium Therapeutics for the treatment of moderate-to-severe acute pain. It utilizes the proprietary Oral Overdose Protected (O2P) technology, a bifunctional prodrug design where the active opioid is released only upon activation by trypsin in the small intestine. This mechanism is intended to provide effective analgesia while preventing abuse via non-oral routes (such as insufflation or injection) and mitigating the risk of fatal overdose from excessive oral ingestion by saturating the activation enzymes. ETR028 has been evaluated in Phase 1 clinical trials to assess its safety, tolerability, and pharmacokinetics compared to standard hydrocodone bitartrate.
