ETX-123 is a preclinical-stage small molecule designed as a potent and selective opener of the voltage-gated potassium channel subunits Kv7.2 (KCNQ2) and Kv7.3 (KCNQ3). Developed by Eliem Therapeutics (now Climb Bio), the compound was intended for the treatment of neuronal excitability disorders, including epilepsy, chronic pain, and depression. In preclinical electrophysiology assays, ETX-123 demonstrated high potency and differentiated selectivity for the Kv7.2/3 heterotetramer. In vivo studies in rat maximal electroshock seizure (MES) models showed dose-dependent inhibition of seizures. Although Phase 1 clinical trials were planned for early 2024, development was halted in July 2023 as the company announced it would explore strategic alternatives.
