Overview

**ETx-22** is a next-generation **antibody-drug conjugate (ADC)** targeting **Nectin-4**, a cell adhesion molecule overexpressed in various solid tumors including urothelial, bladder, triple-negative breast, lung, and ovarian cancers. It comprises a tumor-specific anti-Nectin-4 monoclonal antibody (15A7.5) conjugated via a proprietary β-glucuronide PSARlink™ technology to **exatecan**, a potent **topoisomerase I inhibitor** payload, enabling site-specific release in tumors to enhance efficacy in low Nectin-4-expressing models resistant to enfortumab vedotin (EV) while minimizing skin toxicity. Developed by **Emergence Therapeutics** (acquired by **Eli Lilly**), it shows durable tumor regressions in preclinical PDX models, favorable PK, and a highest non-severely toxic dose >20 mg/kg in nonhuman primates without significant dermal effects, positioning it for advanced solid tumors.[1][2][3][4]
