ETX1153 is a novel, oral diazabicyclooctane (DBO) small molecule inhibitor that specifically targets penicillin-binding protein 3 (PBP3) in *Neisseria gonorrhoeae*. Developed by Entasis Therapeutics (a subsidiary of Innoviva), ETX1153 is designed as a potential treatment for uncomplicated gonorrhea, including infections caused by multidrug-resistant (MDR) and extensively drug-resistant (XDR) strains. Unlike traditional DBOs that primarily function as beta-lactamase inhibitors, ETX1153 exhibits potent intrinsic antibacterial activity by binding to PBP3, thereby inhibiting bacterial cell wall peptidoglycan synthesis and leading to cell death. Its development addresses the urgent need for new oral therapies as resistance to current frontline treatments, such as ceftriaxone, continues to rise globally.
