Overview

ETX1317 is an investigational **small-molecule diazabicyclooctane serine beta-lactamase inhibitor** developed by Entasis Therapeutics and now associated with Innoviva following Entasis' acquisition. It is the active metabolite of the oral prodrug ETX0282 and is designed to inhibit a broad range of serine beta-lactamases, including class A, class C, and class D enzymes such as CTX-M-15, KPC-2, SHV-5, TEM-1, AmpC, P99, and OXA-48. By reversibly covalently inhibiting these resistance enzymes, ETX1317 restores the activity of partner beta-lactam antibiotics against multidrug-resistant Enterobacterales, including carbapenem-resistant strains. Its clinical development has primarily been in the oral combination of ETX0282 with cefpodoxime proxetil for complicated urinary tract infections and related resistant Gram-negative infections rather than as a standalone marketed product.
