This approach aims to restore functional UBE3A protein expression in the brain, addressing the underlying genetic cause of Angelman syndrome with a potentially durable, one-time treatment. Preclinical studies have demonstrated safety, broad biodistribution in non-human primates, and effective upregulation of paternal UBE3A across disease-relevant brain regions[2][7][9]. 2. **ETX Pharma's ETX-201** is an oral small molecule drug developed as a disease-modifying therapy for Crohn's disease and other inflammatory bowel diseases (IBD). It acts as an inhibitor of tumor necrosis factor (TNF) synthesis—specifically targeting TNF1—and also inhibits other inflammatory mediators such as iNOS, IL1, IL6, and p38 MAPK. Unlike biologic anti-TNF antibodies that completely neutralize TNF systemically (and are associated with significant side effects), ETX-201 is designed to act locally within the gastrointestinal tract to reduce inflammation without fully suppressing protective immune responses or causing systemic immunosuppression[1][4][5].
