EU-5346 is a small molecule BH3-mimetic developed by Eutropics Pharmaceuticals that targets anti-apoptotic members of the BCL-2 family, specifically Mcl-1, BCL-2, and BCL-XL. By mimicking the BH3 domain of pro-apoptotic proteins, EU-5346 disrupts the protein-protein interactions (PPIs) that prevent cancer cells from undergoing apoptosis. The compound was primarily investigated for the treatment of hematologic malignancies such as acute myeloid leukemia (AML) and multiple myeloma (MM), where these anti-apoptotic proteins are often overexpressed and drive cell survival. Although Eutropics initially advanced EU-5346 and related compounds like EU-517 through preclinical development with support from NCI-SBIR grants, the company has since transitioned its focus to the PRIMABS-Dx companion diagnostic platform, and its internal drug development programs are no longer active.
