EU307 is a fourth-generation autologous chimeric antigen receptor T cell (CAR-T) therapy developed by Eutilex for the treatment of glypican-3 (GPC3)-positive advanced hepatocellular carcinoma and other solid tumors. It specifically targets the tumor-associated antigen GPC3, which is highly expressed in hepatocellular carcinoma. To enhance efficacy in solid tumors, EU307 incorporates genetic engineering to secrete interleukin-18 (IL-18), which improves CAR-T cell persistence and function within the immunosuppressive tumor microenvironment. This dual mechanism enables both direct cytotoxicity against cancer cells and reprogramming of the tumor microenvironment to support anti-tumor immunity. The drug is currently being evaluated in a first-in-human Phase 1 clinical trial for patients with advanced hepatocellular carcinoma who have failed standard therapies[1][2][4][6].
