Eugenol

Eugenol is a naturally occurring phenolic compound found in several plants such as clove, cinnamon, and bay leaves. It is widely used as a topical antiseptic and analgesic in dental preparations—often combined with zinc oxide for root canal sealing and pain control—and for the temporary relief of toothaches. Eugenol exhibits anti-inflammatory, antioxidant, antimicrobial, antipyretic, neuroprotective, antifungal, and analgesic properties. Its mechanism of action includes inhibition of sodium channel currents (which underlies its local anesthetic effect), positive allosteric modulation of GABA-A receptors (contributing to general anesthetic properties), inhibition of pro-inflammatory mediators like nitric oxide synthase and cyclooxygenase enzymes, scavenging reactive oxygen species (antioxidant effect), and mild monoamine oxidase inhibition in vitro. Although not currently available in any FDA-approved products as a pharmaceutical drug or over-the-counter medication in the US market, it remains present in unapproved OTC dental products for toothache relief[1][3][5][6].
