Euphol is a tetracyclic triterpene alcohol isolated from the sap of plants in the *Euphorbiaceae* family, most notably *Euphorbia tirucalli* (pencil tree). It has demonstrated a broad range of pharmacological activities in preclinical studies, including anti-inflammatory, analgesic, and antineoplastic properties. In the context of neuro-oncology, euphol has shown significant therapeutic potential against high-grade gliomas (HGG), including glioblastoma (GBM) and pediatric HGG. Its mechanism of action involves the induction of apoptosis, inhibition of cell proliferation and migration, and modulation of signaling pathways such as protein kinase C (PKC). Euphol has also been found to sensitize glioma cells to the standard chemotherapy agent temozolomide (TMZ), suggesting its potential as a synergistic combination therapy for aggressive brain tumors. Research is primarily being conducted by academic and clinical institutions in Brazil and Portugal.
