Drug intelligence / Profile preview
Eurolen is a generic formulation of lenalidomide, an immunomodulatory imide drug (IMiD) developed by Eurolab. It is approved in South Africa for the treatment of multiple myeloma. Eurolen acts as a molecular glue that binds to cereblon (CRBN), a component of the CRL4-CRBN E3 ubiquitin ligase complex. This binding alters the substrate specificity of the ligase, leading to the recruitment, ubiquitination, and subsequent proteasomal degradation of the lymphoid transcription factors Ikaros (IKZF1) and Aiolos (IKZF3). The degradation of these transcription factors downregulates interferon regulatory factor 4 (IRF4) and c-Myc, which are essential for the survival of multiple myeloma cells, thereby inducing apoptosis and inhibiting tumor cell proliferation.
