EV-siRNA-41 is an extracellular vesicle (EV)-encapsulated, chemically modified small interfering RNA (siRNA) therapeutic developed by Exollence Biotechnology for the treatment of KRAS G12D-mutant pancreatic ductal adenocarcinoma (PDAC). Utilizing the proprietary shock wave-engineered extracellular vesicle (SWEET™) platform, the drug achieves efficient and stable encapsulation of siRNAs designed for high stability and reduced innate immune activation. EV-siRNA-41 specifically targets and silences the KRAS G12D mutation, leading to the suppression of downstream ERK and AKT signaling pathways. Preclinical studies have demonstrated that EV-siRNA-41 achieves superior tumor accumulation compared to lipid nanoparticles (LNPs) and induces significant tumor regression at microdose levels, outperforming leading small-molecule KRAS G12D inhibitors such as MRTX1133.
