EV20 is a humanized monoclonal antibody that specifically targets HER-3 (receptor tyrosine-protein kinase erbB-3). Its principal mechanism is antagonism of HER-3, inhibiting both ligand-dependent and ligand-independent activation, thereby interrupting downstream oncogenic signaling pathways. EV20 is rapidly internalized by HER-3–expressing tumor cells, making it suitable for use as an antibody-drug conjugate (ADC) platform. In its unconjugated form, EV20 can inhibit tumor growth; as an ADC (e.g., EV20-MMFA, EV20-sss-vc/MMAF, EV20-Sap, EV20/NMS-P945), it is conjugated to cytotoxic agents, allowing efficient and selective delivery of these payloads to HER-3–expressing tumors. EV20-based ADCs have demonstrated potent, specific, and target-dependent antitumor activity in preclinical models of melanoma, breast cancer (including trastuzumab-resistant HER2-positive tumors), and liver cancer, among others[3][1][2][4][5].
