Overview
EV20-Sap is an immunotoxin or antibody-drug conjugate (ADC) composed of the humanized anti-HER3 monoclonal antibody EV20 conjugated to saporin, a type I ribosome-inactivating protein (RIP). Developed by MediaPharma, the agent specifically targets the Human Epidermal Growth Factor Receptor 3 (HER3/ErbB3), which is frequently upregulated in various solid tumors, including melanoma. Upon binding to the extracellular domain of HER3, the complex is internalized via endocytosis. Once inside the cell, saporin is released into the cytoplasm, where it depurinates the 28S ribosomal RNA, effectively halting protein synthesis and inducing apoptosis. EV20-Sap is currently in preclinical development for the treatment of melanoma.
