Evans blue is a synthetic azo dye with a very high affinity for serum albumin, making it useful as a physiological tracer and diagnostic agent. It is primarily used to estimate blood plasma volume, assess vascular and cellular membrane permeability, detect lymph nodes, and localize tumor lesions. Evans blue acts pharmacologically as a negative allosteric modulator of AMPA and kainate receptors, an inhibitor of vesicular glutamate transporters, and an antagonist at P2X-selective purinoceptors. It has also demonstrated dual antiviral effects against hepatitis B virus by inhibiting viral binding to host cells via NTCP and interfering with capsid assembly. Evans blue is non-toxic at diagnostic doses and can be administered intravenously; its strong binding to albumin allows for visualization in tissues using colorimetric or fluorescence-based methods[1][2][3][4][6].
