EvCAR-KHYG-1 is a genetically modified natural killer (NK) cell line derived from the human KHYG-1 cell line. It expresses a chimeric antigen receptor (CAR) targeting epidermal growth factor receptor variant III (EGFRvIII), which is commonly found in glioblastoma and other tumors. The CAR is introduced via lentiviral transduction, conferring specificity for EGFRvIII-expressing tumor cells. In preclinical studies, EvCAR-KHYG-1 shows potent cytotoxic activity, leading to apoptosis and production of Th1-type cytokines (interleukin-2, interferon-γ, and tumor necrosis factor-α) against EGFRvIII-positive glioblastoma cells in vitro. However, in vivo studies suggest it may induce pseudoprogression-like features in tumors rather than consistent inhibition of tumor growth, highlighting the complexity of tumor microenvironment interactions in solid cancers. The base KHYG-1 cell line is notable for its high cytotoxic activity and a point mutation in TP53 (p53), which may affect its biological properties[1][2][5][7][8].
