Overview

EVE16-engineered natural killer cells are an investigational engineered cell therapy being developed by InnDura Therapeutics, utilizing technology licensed from the Dana-Farber Cancer Institute and the Massachusetts Institute of Technology. The therapy involves the expression of a synthetic immune receptor called EVE16 (Enhanced-Valency Engineered CD16A) on natural killer (NK) cells. This receptor is a modified version of the CD16A (FcγRIIIa) receptor, engineered to trigger potent cellular activation through the endogenous CD3ζ adaptor signaling protein. The EVE16 platform is designed to enhance anti-tumor efficacy by enabling both direct CAR-like cytotoxicity against tumor antigens and robust antibody-dependent cellular cytotoxicity (ADCC). A key feature of the EVE16 design is its ability to minimize trogocytosis and tonic signaling, which are common factors leading to cell exhaustion in other engineered therapies. Preclinical studies of CD19-targeted EVE16 memory-like NK cells have shown significant expansion, persistence, and leukemic clearance in animal models, supporting its development for both hematologic malignancies and solid tumors.
