A combination therapy consisting of the mTOR inhibitor everolimus and the standard chemotherapy agents cytarabine and daunorubicin. This regimen is being evaluated for the treatment of relapsed acute myeloid leukemia (AML). Everolimus acts as a chemosensitizer by inhibiting the mammalian target of rapamycin (mTOR), a pathway often activated in AML that promotes cell survival and resistance to chemotherapy. Cytarabine is an antimetabolite that inhibits DNA synthesis, while daunorubicin is an anthracycline that causes DNA damage through intercalation and inhibition of topoisomerase II. The combination is designed to enhance the efficacy of the standard "7+3" induction backbone in patients who have failed prior therapy.
