Everolimus + ranibizumab is an investigational combination therapy consisting of two distinct agents with complementary mechanisms of action. Everolimus is an oral small molecule inhibitor of the mammalian target of rapamycin (mTOR), a key regulator in cell growth, proliferation, and angiogenesis. Ranibizumab is a recombinant humanized monoclonal antibody fragment that binds to and inhibits vascular endothelial growth factor A (VEGF-A), thereby suppressing abnormal blood vessel formation and vascular permeability. The rationale for combining these agents lies in their potential synergistic effects on inhibiting both mTOR-mediated tumor cell survival/proliferation and VEGF-driven angiogenesis, particularly relevant in diseases characterized by pathological neovascularization such as certain cancers or retinal disorders.
