This is a combination regimen consisting of three drugs—everolimus, trastuzumab, and vinorelbine—used primarily in the treatment of HER2-positive advanced or metastatic breast cancer, particularly in patients who are resistant to trastuzumab and have been pre-treated with a taxane. Everolimus is an oral small molecule inhibitor of mammalian target of rapamycin (mTOR), which blocks cell growth and proliferation signaling pathways. Trastuzumab is a monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2), inhibiting HER2-mediated signaling and inducing antibody-dependent cellular cytotoxicity. Vinorelbine is a small molecule chemotherapeutic agent that inhibits microtubule assembly, disrupting mitosis. The combination aims to overcome resistance to anti-HER2 therapy by simultaneously inhibiting mTOR-driven survival pathways and HER2 signaling while providing cytotoxic chemotherapy[1][3][4][5][7].
