Evexomostat is a synthetic copolymer-drug conjugate of a fumagillin-derived methionine aminopeptidase 2 (METAP2) inhibitor, conjugated to the biocompatible polymer poly(N-(hydroxypropyl)methacrylamide) (HPMA). It is being developed primarily for the treatment of cancers, especially in patients who are overweight or have systemic metabolic dysfunction. The drug acts by inhibiting METAP2, leading to cell cycle arrest and inhibition of tumor angiogenesis. Preclinical and clinical data suggest it may also improve metabolic disorders such as hyperglycemia and obesity in cancer patients. Evexomostat is under investigation in combination with standard chemotherapy agents like eribulin for triple-negative breast cancer (TNBC), particularly in populations with metabolic comorbidities[1][5][6][7][8].
