Evixapodlin is a small-molecule, orally bioavailable inhibitor of programmed cell death-ligand 1 (PD-L1), developed for the treatment of cancer and previously investigated for hepatitis B. It works by dimerizing PD-L1, forming a trimer complex that blocks the binding site for PD-1 on T cells, thereby inhibiting the PD-1/PD-L1 interaction both at the cell surface and intracellularly. This mechanism prevents tumor cells from evading immune detection and enhances T-cell mediated immune responses. Evixapodlin is particularly potent in cells with high PD-L1 expression, such as tumor cells, while sparing normal tissue with lower expression levels. The drug has demonstrated anticancer and antiviral activities in preclinical studies and has been evaluated in phase I/II clinical trials for solid tumors[3][5][7].
