EVM15 is an internally developed small molecule inhibitor of ubiquitin-specific protease 1 (USP1), currently in preclinical development by Everest Medicines (also known as Genting Pharmaceutical in certain regions) for the treatment of oncology indications, particularly solid tumors. USP1 is a deubiquitinating enzyme that plays a critical role in the DNA damage response (DDR) by regulating the Fanconi anemia pathway and translesion synthesis. By inhibiting USP1, EVM15 aims to disrupt DNA repair mechanisms, potentially leading to synthetic lethality in tumors with specific genetic vulnerabilities, such as those deficient in BRCA1/2, or enhancing the efficacy of other DNA-damaging therapies like PARP inhibitors. The program is currently in the Pre-IND stage as part of Everest Medicines' strategic expansion into proprietary drug discovery.
