Overview

EVM401 refers to a series of novel small-molecule compounds developed by Enveric Biosciences, designed as non-hallucinogenic derivatives of mescaline and tryptamine. These compounds are engineered to act as neuroplastogens—agents that promote neuroplasticity in the brain—while minimizing or eliminating hallucinogenic effects. The EVM401 series includes carboxylated derivatives of tryptamine-based and mescaline-derived molecules, specifically Cl-substituted isopropylamine fused heterocyclic mescaline derivatives and related phenylalkylamines/indolethylamines. Their mechanism involves modulation of multiple key neurological receptors implicated in mental health disorders, including the alpha-2A adrenergic receptor (ADRA2A), serotonin 5-HT2C receptor, dopamine transporter (DAT), and potentially serotonin 5-HT2A receptor. The primary indications under development are depression, anxiety disorders, addiction/substance use disorders, opioid withdrawal management, and ADHD. Preclinical data suggest these agents can promote clinically relevant neuroplasticity without inducing hallucinations—a major limitation for traditional psychedelic therapies—potentially enabling broader outpatient use for mental health conditions[1][3][4][5][6][7].
