EVO101

EVO101 is a novel small molecule inhibitor of interleukin 1 receptor-associated kinase 4 (IRAK4), developed by Evommune for the treatment of inflammatory skin diseases. Its mechanism of action involves inhibition of IRAK4, a key protein in innate immune responses that mediates signaling through Toll-like receptors and IL-1 family cytokines. By blocking IRAK4 activity, EVO101 broadly suppresses both innate and adaptive inflammation. The drug was primarily developed as a topical cream for conditions such as atopic dermatitis and rosacea. Clinical studies have also explored its use in hidradenitis suppurativa. Despite promising preclinical data and early-phase clinical trials showing good tolerability, development for atopic dermatitis and rosacea has been discontinued following phase 2 results[2][5][6][7][8].
