Drug intelligence / Profile preview
EVO570 (formerly HC118, known as HP570 in Greater China) is an investigational, orally administered molecular glue degrader (MGD) of Vav guanine nucleotide exchange factor 1 (VAV1). Discovered by Hinova Pharmaceuticals and licensed to Evommune for development outside Greater China, EVO570 is designed to selectively target and degrade VAV1, a signaling protein largely restricted to immune cells that transmits signals from T-cell and B-cell receptors to drive activation, cytokine production, and migration. By recruiting the cereblon (CRBN) E3 ubiquitin ligase, EVO570 directs VAV1 to the cell's proteasomal machinery for degradation, thereby quieting the immune activity underlying chronic inflammatory and autoimmune diseases. In preclinical studies, EVO570 demonstrated sub-nanomolar potency, high selectivity, and robust disease control in models of rheumatoid arthritis, inflammatory bowel disease, and experimental autoimmune encephalomyelitis. The US FDA has accepted an Investigational New Drug (IND) application for EVO570, enabling the initiation of a Phase 1 trial evaluating safety, pharmacokinetics, and target engagement, with a Phase 2 trial in rheumatoid arthritis planned for 2027.
