Overview

Evogliptin is a potent, highly selective, and long-acting small molecule inhibitor of dipeptidyl peptidase-4 (DPP-4) developed by Dong-A ST for the treatment of type 2 diabetes mellitus. By inhibiting the DPP-4 enzyme, it prevents the degradation of incretin hormones such as glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP), thereby enhancing glucose-dependent insulin secretion and suppressing glucagon release. Evogliptin is characterized by its high potency (low EC50) and a pharmacokinetic profile that supports once-daily dosing. It has been approved in several countries, including South Korea, Brazil, and India, and is used both as monotherapy and in combination with other antidiabetic agents like metformin and dapagliflozin. Beyond its primary metabolic indication, research has explored its potential protective effects in renal fibrosis, diabetic cardiomyopathy, and as a synergistic therapy for glioblastoma.
