EVOLVE-106 is a preclinical-stage, multifunctional T cell engager with integrated CD2 costimulation, developed by EvolveImmune Therapeutics as a precision immunotherapy for solid tumors, especially estrogen and HER2 receptor low breast cancer. It is designed to selectively target the immune checkpoint protein B7-H4 (V-set domain-containing T-cell activation inhibitor 1, VTCN1), while simultaneously activating CD2 on T cells to amplify their tumor killing function, overcome low tumor immunogenicity, conditionally activate adaptive immunity, and limit T cell dysfunction. Preclinical data demonstrate robust single-agent anti-tumor activity superior to traditional B7-H4 x CD3 T cell engagers. EVOLVE-106 is intended for solid tumors where low immunogenicity and immune escape represent significant clinical challenges. First-in-human trials are anticipated following preclinical success.
