EVOLVE104 is a first-in-category, trispecific T cell engager (TriTE) designed for the treatment of solid tumors. It uniquely integrates CD2 co-stimulation with CD3 engagement to enhance and sustain T cell effector function against cancer cells. The molecule conditionally targets ULBP2/5/6 antigens—members of the UL16 binding protein family that are highly expressed in squamous and select other malignancies (such as non-small cell lung cancer, head and neck cancers, and muscle-invasive bladder cancer), but have restricted expression in normal tissues. By combining an affinity-tuned CD3 binder with a CD2-selective fusion protein, EVOLVE104 delivers coordinated costimulatory signals to T cells upon tumor recognition. Preclinical studies demonstrate potent tumor-killing activity superior to standard chemotherapies (cisplatin/gemcitabine) in ULBP2-expressing models, favorable pharmacokinetics, absence of cytokine release syndrome or off-target toxicity at efficacious doses in animal models, and strong rationale for combination with anti-PD-1 therapies like pembrolizumab[1][3][4][5][6].
