This combination therapy regimen consists of evorpacept (ALX148), pembrolizumab, and pegylated liposomal doxorubicin (PLD). It is currently being evaluated in a Phase II clinical trial for the treatment of recurrent platinum-resistant ovarian cancer. Evorpacept is a next-generation CD47 blocker designed as a fusion protein that binds CD47 with high affinity while utilizing an inactive human immunoglobulin Fc region to minimize hematologic side effects. Pembrolizumab is a programmed cell death protein 1 (PD-1) inhibitor that enhances T-cell mediated anti-tumor activity. Pegylated liposomal doxorubicin is a cytotoxic chemotherapy agent that inhibits DNA topoisomerase II. The combination aims to leverage the synergy between innate immune activation (via CD47 blockade), adaptive immune checkpoint inhibition (via PD-1 blockade), and direct cytotoxic tumor cell killing to overcome resistance in ovarian cancer.
