Overview

This is a combination therapy consisting of **evorpacept** (ALX148), **trastuzumab** (Herceptin), and **gemcitabine** (Gemzar). Evorpacept is a high-affinity CD47-blocking fusion protein designed to enhance immune-mediated cancer cell elimination by blocking the "don't eat me" signal from CD47, thereby promoting phagocytosis of tumor cells. Trastuzumab is a monoclonal antibody targeting human epidermal growth factor receptor 2 (HER2/ERBB2) to inhibit proliferation of HER2-overexpressing tumor cells and mediate antibody-dependent cellular cytotoxicity (ADCC). Gemcitabine is a deoxycytidine analog antimetabolite which incorporates into DNA during replication, causing masked DNA chain termination and apoptosis in dividing cancer cells. This triple combination is under investigation for HER2-positive metastatic breast cancer and other advanced solid tumors, aiming to synergistically promote tumor cell death through immune and cytotoxic mechanisms[7][10][3][9][6].
