rejuvenates the anti-tumor activity of innate immune cells by blocking CD47 to enhance phagocytosis. Trastuzumab (Herceptin) is a monoclonal antibody that targets the HER2 receptor, inhibiting the proliferation of HER2-overexpressing tumor cells. Paclitaxel is a cytotoxic small molecule that stabilizes microtubules, arresting cell division. This combination acts synergistically by combining immune checkpoint blockade (evorpacept), targeted inhibition of tumor growth (trastuzumab), and cytotoxic chemotherapy (paclitaxel). It is being developed primarily for gastric and gastroesophageal junction cancers after progression following prior anti-HER2 therapy, based on results from phase 2/3 ASPEN-06 clinical trial, where it has shown improved overall response rates and duration of response compared to control arms[1][2][3][4].
