A four-drug combination therapy consisting of **evorpacept** (a CD47-blocking fusion protein), **trastuzumab** (a monoclonal antibody against HER2), **ramucirumab** (a monoclonal antibody antagonist of VEGFR-2), and **paclitaxel** (a microtubule-stabilizing chemotherapy). Each agent contributes via a distinct mechanism with the goal of synergistic antitumor activity: Evorpacept promotes antibody-mediated phagocytosis and enhances immune response; trastuzumab inhibits HER2 signaling and mediates immune effector functions; ramucirumab blocks angiogenesis by targeting VEGFR-2; and paclitaxel disrupts microtubule function to inhibit cell division and induce apoptosis. This combination is under investigation primarily in HER2-positive, advanced or metastatic gastric or gastroesophageal junction (GEJ) cancer.
