EVP-0962 is a proprietary small molecule drug developed as a gamma-secretase modulator (GSM) for the treatment of Alzheimer's disease and mild cognitive impairment. Unlike gamma-secretase inhibitors, EVP-0962 selectively modulates the activity of gamma-secretase to reduce the production of toxic amyloid-beta 42 (Aβ42) peptides without affecting other essential substrates such as Notch. This selective modulation is intended to slow disease progression by lowering Aβ42 levels and amyloid plaque formation, while minimizing safety risks associated with broad inhibition of gamma-secretase. Preclinical studies demonstrated that EVP-0962 reduced Aβ1–42 peptide levels, decreased amyloid plaque buildup, and reversed behavioral deficits in transgenic mouse models of Alzheimer’s disease. Clinical development reached Phase 2 trials evaluating safety, tolerability, pharmacokinetics, and effects on cerebrospinal fluid amyloid concentrations in healthy volunteers and patients with mild cognitive impairment or early Alzheimer’s disease[1][5][6].
