Drug intelligence / Profile preview
EVsTIM3OE (EVs^TIM3OE) is a preclinical, engineered extracellular vesicle (EV) product designed to overexpress T-cell immunoglobulin and mucin domain-containing protein 3 (TIM3, encoded by HAVCR2). Developed by researchers at the Shanghai Ninth People's Hospital and Shanghai Jiao Tong University School of Medicine, these cell-derived nanotherapeutics are engineered from plasmid-transfected bone marrow stromal cells (BMSCs). EVsTIM3OE is designed to target macrophages, promoting their reprogramming from a pro-inflammatory to an anti-inflammatory M2-like phenotype. Mechanistically, the overexpressed TIM3 on the EVs inhibits the p38/MAPK signaling pathway to suppress inflammation and release anti-inflammatory cytokines (such as IL-10 and TGF-β1), while also activating the Bmp2 promoter to enhance macrophage BMP2 secretion, thereby promoting osteogenesis and osseointegration. For therapeutic delivery, EVsTIM3OE is loaded into a gelatin-methacryloyl (GelMA) hydrogel for sustained release, with potential applications in large craniomaxillofacial bone defect repair and refractory wound healing.
