EVT-3023 (also known as EVT-0003023) is a preclinical small-molecule inhibitor of PKMYT1 being developed by Evariste Ltd., an AI-enabled drug discovery company based in London. PKMYT1 is a kinase that regulates the cell cycle, specifically the G2/M transition, and is frequently upregulated in multiple cancers. EVT-3023 is designed to exploit synthetic lethality, particularly in tumors with genomic vulnerabilities such as CCNE1 amplification. The compound has demonstrated efficacy and tolerability in multiple tumor models and is reported to possess best-in-class selectivity for PKMYT1 over related kinases like WEE1. This selectivity is intended to widen the therapeutic window and minimize toxicities associated with competing PKMYT1 inhibitor programs. Additionally, Evariste has identified and validated a novel biomarker for PKMYT1 that may help identify and expand the eligible patient population for this therapy. The drug is currently in preclinical development for oncology indications and is scheduled for presentation at the AACR Annual Meeting 2025.
