Overview

EVT 401 is an orally administered small molecule antagonist of the purinergic P2X7 receptor, originally developed by Renovis and further advanced by Evotec for inflammatory and immunological disorders, with rheumatoid arthritis as an initial lead indication.[1][9] By selectively blocking the ATP-gated P2X7 ion channel on immune cells, EVT 401 inhibits ATP-stimulated interleukin-1β release and other downstream proinflammatory cytokine pathways, providing a potential disease-modifying approach for conditions driven by chronic inflammation and neuroinflammation.[1][5][6][7][9] The drug has completed first-in-human Phase I studies demonstrating good safety, tolerability, and pharmacodynamic target engagement after single oral dosing, and is being further developed in China by Zhejiang Conba Pharmaceutical under license from Evotec.[1][3][5][9][11]
