Ex-vivo expanded γδ T lymphocytes are a form of adoptive cell therapy in which γδ T cells are isolated from peripheral blood (either autologous or allogeneic), then activated and proliferated outside the body using specific stimulants such as zoledronate and interleukin-2 (IL-2), artificial antigen-presenting cells (aAPCs), or other cytokines. These cells are then reinfused into patients as an immunotherapy for cancer and other diseases. Unlike conventional αβ T cells, γδ T lymphocytes recognize antigens in a major histocompatibility complex-independent manner, allowing them to target a broad range of tumor-associated antigens with innate cytotoxicity while minimizing the risk of graft-versus-host disease. Ex-vivo expansion protocols can achieve large-scale production under GMP-like conditions for clinical use. The primary indications under investigation include hematologic malignancies such as acute myeloid leukemia and solid tumors like neuroblastoma and glioblastoma[1][2][4][6][7].
