Ex-vivo expanded γδ T lymphocytes are an adoptive cell therapy involving the isolation, activation, and expansion of gamma delta (γδ) T cells outside the body before re-infusion into a patient. Unlike conventional αβ T cells, γδ T cells recognize antigens in a Major Histocompatibility Complex (MHC)-independent manner, often through the TCR or natural killer receptors like NKG2D. This allows them to target a broad range of tumors without the strict requirement for HLA matching, reducing the risk of Graft-versus-Host Disease (GvHD) in allogeneic settings. In the context of relapsed or refractory acute myeloid leukemia (AML), these cells are typically expanded from healthy donors (allogeneic) and administered to harness their potent innate-like anti-leukemic cytotoxicity. The specific program described is being evaluated in Phase 1 clinical trials by academic institutions in China, including Union Hospital of Tongji Medical College.
