Ex vivo-expanded HER2-specific T cells is an investigational adoptive cell therapy (ACT) developed by the University of Washington for the treatment of advanced HER2-positive breast cancer. The therapeutic approach involves a multi-step process: first, the patient is primed in vivo with a HER-2/neu peptide vaccine to stimulate the production of HER2-specific memory CD8+ T cells. These cells are then harvested from the patient via leukapheresis, expanded ex vivo in a laboratory setting to increase their numbers, and re-infused intravenously. Prior to infusion, patients receive lymphodepleting chemotherapy with cyclophosphamide to create a favorable environment for T-cell engraftment and persistence. The infused T cells are designed to specifically recognize and eliminate cancer cells that overexpress the Human epidermal growth factor receptor 2 (HER2/neu) protein.
