Ex vivo produced oral mucosal composites (EVPOMCs) are tissue-engineered autologous grafts developed for oral cavity reconstruction. The process involves harvesting a small biopsy of oral mucosa from a patient, isolating the epithelial cells (keratinocytes) and fibroblasts, and expanding them in a laboratory setting. These cells are typically seeded onto a scaffold—often a human cadaveric dermis (AlloDerm) or a collagen-based matrix—to create a multilayered, three-dimensional tissue structure that mimics the native oral mucosa. Once matured ex vivo, the composite is grafted back into the donor patient to provide a source of keratinized or attached mucosa. This technology aims to overcome the limitations of traditional grafts, such as limited donor site availability and the significant pain associated with palatal tissue harvesting.
