**EX101** is a proprietary buffered effervescent tablet formulation of alendronate sodium, a bisphosphonate administered once weekly for the treatment of postmenopausal osteoporosis in women and to increase bone mass in men with osteoporosis. It acts as a specific inhibitor of osteoclast-mediated bone resorption by binding to hydroxyapatite in bone and inhibiting farnesyl pyrophosphate synthase in osteoclasts, thereby reducing bone turnover and increasing bone mineral density. Developed by EffRx Pharmaceuticals to improve gastric tolerance compared to standard alendronate tablets, EX101 rapidly clears the esophagus, maintains higher gastric pH during emptying, and minimizes mucosal irritation; its NDA was accepted by the FDA.[1][3][4][5]
