Overview

Exenatide-XTEN is a long-acting recombinant fusion protein designed for the treatment of type 2 diabetes mellitus. It consists of the glucagon-like peptide-1 (GLP-1) receptor agonist exenatide fused to an XTEN polymer. XTEN is a non-repetitive, hydrophilic, and biodegradable polypeptide technology developed to extend the half-life of therapeutic peptides by increasing their hydrodynamic radius, thereby reducing renal clearance. As a GLP-1 receptor agonist, exenatide-XTEN mimics the action of endogenous GLP-1, stimulating glucose-dependent insulin secretion, inhibiting glucagon release, and slowing gastric emptying. The fusion protein was engineered to allow for significantly less frequent dosing (e.g., once-monthly) compared to standard exenatide formulations. Development was primarily conducted by Amunix in collaboration with Versartis.
