Overview

Exendin-asp3 (ExD3) is a modified peptide agonist of the glucagon-like peptide-1 receptor (GLP-1R), derived from the structure of exendin-4. It is characterized as a biased agonist that specifically promotes rapid internalization of the GLP-1R into endosomes. Despite its high propensity for driving receptor trafficking and its accumulation in endocytic compartments, research indicates that exendin-asp3 is a relatively inefficient driver of cAMP production in pancreatic β-cells and insulin secretion from the pancreas compared to other GLP-1R agonists. It is primarily utilized as a pharmacological tool in research to investigate the spatial distribution of GPCR signaling and the therapeutic implications of biased agonism in the treatment of type 2 diabetes and obesity.
