Overview

Exenokine-21 is a recombinant fusion protein comprising interleukin-21 (IL-21) fused to an anti-human serum albumin (HSA) single-domain nanobody. It is designed as a half-life extended IL-21 agonist for cancer immunotherapy. Exenokine-21 acts as an IL-21 receptor agonist and has demonstrated superior antitumor activity compared to unmodified IL-21 in preclinical models. The drug enhances immune responses by activating pathways associated with CD8+ T cell and NK cell cytotoxicity and can potentiate the efficacy of immune checkpoint inhibitors such as PD1 and TIGIT blockades. It is being developed primarily for use in combination with standard-of-care therapies for various cancers including lymphoma, head and neck cancer, renal cell carcinoma (RCC), melanoma, solid tumors, and liver cancer[1][2][3][4].
