Exlinkibart (LVGN6051) is a humanized monoclonal antibody that acts as a conditional agonist of CD137 (also known as 4-1BB), designed for selective activation in the tumor microenvironment through Fcγ-receptor IIB binding. This mechanism enables targeted immune activation, enhancing antitumor T cell and natural killer cell responses while minimizing systemic toxicity. Exlinkibart is developed using Lyvgen Biopharma’s xLinkAb platform technology and has demonstrated potent antitumor activity both as monotherapy and in combination with anti-PD-1 antibodies such as pembrolizumab. It is being investigated primarily for advanced solid tumors, including melanoma, non-small cell lung cancer (NSCLC), gastrointestinal malignancies, head and neck squamous cell carcinoma (HNSqCC), sarcoma, esophageal cancer, hepatocellular carcinoma, ovarian cancer, soft tissue cancers, uveal melanoma, and lymphoma.
