Overview

EXN407 is a first-in-class, twice-daily topical small molecule inhibitor of serine/arginine-rich protein-specific kinase 1 (SRPK1), developed as an eye drop for the treatment of retinal vascular diseases such as non-proliferative diabetic retinopathy (NPDR) and diabetic macular edema (DME). By inhibiting SRPK1, which regulates the alternative splicing of vascular endothelial growth factor (VEGF), EXN407 selectively targets pro-angiogenic VEGF isoforms responsible for abnormal blood vessel growth and leakage in the retina. Clinical studies have demonstrated that EXN407 is safe and well-tolerated with promising biological activity—showing sustained decreases in macular thickness and significant reductions in vascular leakage compared to placebo. The drug aims to provide a non-invasive alternative to anti-VEGF injections for patients with diabetic eye disease[2][3][5][7].
