CDK-003 (also known as exoIL-12 or exoInterleukin-12) is an engineered exosome-based therapy developed by Codiak BioSciences for the treatment of cutaneous T-cell lymphoma (CTCL) and other solid tumors. The drug consists of an exosome displaying single-chain interleukin 12 (IL-12), a pro-inflammatory cytokine, on its surface. This design enables targeted activation of immune effector cells such as T cells and natural killer (NK) cells via engagement with the IL-12 receptor. By localizing IL-12 delivery to tumor sites through intralesional administration, CDK-003 aims to stimulate anti-tumor immunity while minimizing systemic toxicity commonly associated with systemic IL-12 therapies. Clinical development has focused primarily on CTCL; previous studies have shown that localized delivery can improve or clear tumors without the severe side effects seen with intravenous IL‑12[1][2][3][4][5].
