Drug intelligence / Profile preview
Exosome-SIRPα (also known as SBI-101) is an engineered exosome-based cancer immunotherapy developed by ShiftBio. It is designed to display signal-regulatory protein alpha (SIRPα) on the exosomal membrane using ShiftBio's proprietary Maxisome platform. By presenting SIRPα, the exosomes target and bind to CD47 (the 'don't eat me' signal) overexpressed on tumor cells. This interaction blocks the CD47-SIRPα signaling pathway, preventing immune evasion and significantly enhancing the phagocytosis of cancer cells by macrophages and dendritic cells. SBI-101 is being developed for the treatment of solid tumors, particularly those resistant to PD-1 blockade.
