EXP012 is a trispecific antibody T cell engager being developed by Excalipoint Therapeutics for the treatment of gastrointestinal cancers, including colorectal, gastric, and pancreatic cancers. The molecule is designed to simultaneously bind to Cadherin-17 (CDH17), which is overexpressed in gastrointestinal tumors, and the immune cell targets CD3 (on T cells) and 4-1BB (a co-stimulatory receptor). By engaging these three targets, EXP012 aims to recruit and activate T cells specifically within the tumor microenvironment to enhance anti-tumor immunity. The drug was licensed from Lepu Biopharma and is currently in preclinical development, with clinical trials anticipated to begin in 2026.
